Abcore offers immunization of llamas and alpacas, resulting in single domain antibody VHH phage libraries which are used for single domain antibody discovery. Abcore most frequently uses immunized llamas and immunized alpacas and isolates the PMBC cells during immunization for library construction. RNA is used to synthesize cDNA, and the single domain antibody VHH sequences are then generated using Abcore’s proprietary primers. Our proprietary PCR primers ensure the maximum coverage of VHH repertoires. We have performed extensive bioinformatics research on VHH genes and designed novel primers for RT-PCR to amplify VHH cDNA. VHH libraries constructed with these primers have much higher diversity and larger coverage of the VHH repertoires. In fact, we have identified several pico-molar affinity VHH binders that have unique sequences and would have been missed if published primers were used.
Immunized antibody phage libraries are used for the following services:
- Custom llama single domain antibody discovery
- Custom alpaca single domain antibody discovery
